No, you are not technically required to kill each and every monster you encounter. You simply need to kill enough enemies to satisfy the criteria in each area. The only exception to this is Snowdrake, which you must kill in order to progress with the Genocide Route. Sparing or running away from Jerry will not abandon your playthrough.
That's just the way that the game was programmed. When you enter Snowdin and start killing monsters, if you check at the SAVE points you may notice the message that says "That comedian..." If you fail to kill Snowdrake before exhausting the kill counter, the message will change to "The comedian got away," and you will be shunted onto the Neutral Route.
Why Snowdrake specifically, though? I have no idea. You'd have to ask Toby that.
You have to kill snowdrake to activate chilldrake. There is only 1 snowdrake in the intire game, infinite chilldrakes, 1 snowdrakes father, and one (now amalgamated) snowdrakes mother. You can only fight snowdrake and chilldrake, and you can only encounter chilldrake after you kill snowdrake. One is always alive (father), one is amalgamated (mother), one is infinite (chilldrake), and one can only be killed once before being replaced by his/her (no making an ass-out-of-u-and-me-ing genders here!) brother/sister (again, no making an ass-out-of-u-and-me-ing genders here!) forever!
